What is sperm DNA fragmentation

Sperm DNA fragmentation testing helps estimate the proportion of sperm in the ejaculate that have damaged DNA. Even sperm that appear normal at first glance may have fragmented DNA, which can make conception more difficult or affect the further course of pregnancy.

How the test is performed

At our clinic, we perform DNA fragmentation testing using the SCD method. Special chemical substances are used to loosen and visualise the DNA in sperm. If the DNA is undamaged, a characteristic “halo effect” forms around the sperm head.

This test therefore shows what percentage of sperm in the sample have damaged DNA. This information helps the doctor choose the next treatment steps more precisely.

Who the test is suitable for

Sperm DNA fragmentation testing is particularly useful in cases of repeated treatment failures or recurrent spontaneous miscarriages. A major advantage is that no additional sample collection is needed – the test can be performed from the same ejaculate sample as the standard semen analysis.
